The ZytoLight ® SPEC FGFR2/CEN 10 ual Color Probe is designed for the etection of FGFR2 gene amplifications requently observed in breast cancer as ell as in gastric cancer.
The FGFR2 (fibroblast growth factor gene , a.k.a. BEK) gene is located on chromosome 0q26 and encodes splice variants f the receptor tyrosine kinases FGFR2b nd FGFR2c.
Amplification of the FGFR2 gene leads to verexpression of the FGFR2 protein and ubsequently to signal activation. Additionally,during the amplification process the -terminal deletion of FGFR2 can occur ue to exclusion of the last exon from he FGFR2 amplicon. Both, overexpression nd deletion of the last exon result n FGFR2 signaling activation based on onstitutive phosphorylation of the FRS2 daptor molecule.
The process of ligand independent GFR2 signaling leads to a more severe alignant phenotype of these tumors.Moreover, high FGFR2 expression is orrelated with poor overall survival (OS)and poor disease-free survival (DFS) rates n breast cancer patients. Consequently,FGFR2 gene amplification detected by luorescence in situ hybridization might be sed as prognostic marker e.g. in breast ancer.

The SPEC FGFR2/CEN 10 Dual Color robe is a mixture of an orange fluorochrome irect labeled CEN 10 probe pecific for the alpha satellite centromeric egion of chromosome 10 (D10Z1) and a reen fluorochrome direct labeled SPEC GFR2 probe specific for the FGFR2 gene t 10q26.
结果
In a normal interphase nucleus, two orange nd two green signals are expected.Nuclei with amplification of the FGFR2 ene locus 10q26, or aneuploidy of hromosome 10 will show multiple copies f the green signal or large green signal lusters.
